Latar belakang: Aktivitas radikal bebas pada membran sel dapat merusak membran, yang dapat dicegah oleh antioksidan. Likopen dan leukoselektfitosom yang diekstraksi dari biji tomat dan biji anggur berperan sebagai antioksidan pada stres oksidatif akibat sonolisis. Aktivitas antioksidan terhadap spesies oksigen reaktif belum diketahui. Tujuan: Membandingkan aktivitas antioksidan kombinasi likopen+leukoselekfitosom dengan kombinasi beta karoten+vitamin C+vitamin E+selenium pada kerusakan membran eritosit akibat stres oksidatif oleh H2O2. Subyek dan metode: Dengan rancangan randomized clinical trial, relawan dibagi menjadi 3 kelompok; Kelompok A mendapat suplementasi likopen 2,5 mg+leukoselekfitosom 25 mg, kelompok B mendapat beta karoten 6 mg+vitamin C 250 mg+vitamin E 200 mg+selenium 30 mcg, dan kelompok C mendapat plasebo selama 30 hari. Eritrosit para relawan diisolasi, dipajani dengan 500 mM H2O2 selama 10 menit, dan hemoglobin supernatan diukur dengan spektrofotometer 546 nm. Perbedaan rerata densitas optis (DO) supernatan diuji dengan Uji Kruskall-Wallis. Hasil: Rerata DO supernatan pada kelompok A=0,08±0,03; B=0,07±0,02 dan C=0,14±0,07. Terdapat perbedaan bermakna rerata DO supernatan kelompok A dan B terhadap plasebo (p<0,05). Tidak terdapat perbedaan DO supernatan antara kelompok A dan B (p>0,05) Kesimpulan: Kombinasi likopen dan leukoselekfitosom menunjukkan aktivitas antioksidan yang setara dengan kombinasi beta karoten, vitamin C, vitamin E, dan selenium.

Background: Free radical activities on cell membranes lead to membrane destruction and their activities might be inhibited by antioxydants. Lycopene and leucoselectphytosome extracted from tomato and grape seeds have antioxydant activities on oxydative stress induced by sonolysis. Their activities against reactive oxygen species is not well known. Aim: To compare antioxydant activities between combined lycopene+ leucoselectphytosome with combined beta carotene+vitamin C+vitamin E+selenium, on erythrocyte membrane damage due to oxydative stress by H2O2. Method: A randomized clinical trial design was conducted among 3 groups (A: 2,5mg-lycopene+25mg-leucoselectphytosome, B: 6 mg-beta carotene+250 mg-vitamin C+200 mg-vitamin E+30 mcg-selenium, and C:placebo) for 30 days. Erythrocytes were isolated, exposed with 500 mM H2O2 for 10 minutes; Hemoglobin supernatant was measured with 546 nm-spectrophotometer. The mean values of supernatant optical density (SOD) among 3 groups were analyzed with Kruskall-Wallis test. Results: The mean values of SOD on groups A=0.08±0.03; B=0.07±0.02 and C=0.14±0.07. There were significant differences (p<0.05) of SOD between group A as well as group B with group C, but there was no significant differences (p>0.05) of SOD between group A and B. Conclusion: Combination of lycopene+leucoselectphytosome has an antioxydant activity which is similiar to combination of betacarotene + vitamin C + vitamin E + selenium.
